Sample Requirements

Users should be able to prepare their cryo-lamellae or biological samples in cryo-frozen EM grids in their own laboratory. A maximum of 4 grids will be loaded into Krios. In order to ensure efficient use of microscope time, users will be asked to provide TEM grid atlases displaying clearly the lamellae for cellular cryo-ET and higher magnification photographs showing the biological sample for macromolecular cryo-ET. Grids of insufficient quality will be rejected.

Safety level of samples:                                                                                                                                                All BSL1 samples under French regulation will be accepted for cryo-ET experiments.       

BSL2 samples (including any human cell lines) are accepted only if they are chemically fixed (ex. Paraformaldehyde) so that they no longer present any biological risk. Apart from this, it is obligatory to submit an authorization for import / export of human samples and derived products to the Ministry (file to be completed by the user and the ESRF will have to correct and send it). Please plan ahead and get in touch with us early enough because the process will take long time—minimum three months.
