X-ray imaging techniques at third-generation synchrotron radiation facilities add a unique set of capabilities to biological research. Alternative contrast mechanisms and spectroscopic methods have been developed in the past decade, particularly for X-ray microscopy. These new techniques can provide both a reduction of the necessary radiation dose and/or valuable complementary information about the nature of the sample (density, chemical composition, chemical states, structure…) mapped in two or, increasingly, in three dimensions. A major factor in the development of synchrotron-based X-ray imaging is the use of the coherence of the synchrotron beams, which allows higher sensitivity and promises higher spatial resolution. These techniques fully exploit the brilliance of the modern synchrotron sources and take advantage of the improvements of detectors and of new algorithms. The aim of this course is to provide young scientists with an overview of the relevant X-ray imaging techniques available at a synchrotron for multiscale investigations in the biological field, through a combination of technical information, scientific presentations, tutorials, and practicals on beamlines.
